Techniques of leaf infiltration divided according to the pressure used. Spontaneous infiltration is possible under normal atmospheric pressure if the infiltration fluids have sufficiently low surface tension and viscosity. Forced infiltration is driven by the pressure difference generated between the leaf surface and its intercellular spaces. The pressure difference can be generated by exposing target tissues to the vacuum conditions. For this purpose, a vacuum chamber with a pump connected to it can be used, or a suitably large syringe in which the infiltrated leaves or their fragments are placed and then subjected to the vacuum by pulling on the plunger. The infiltration fluid can also be directly pressed inside the leaf using a needleless syringe, the outlet of which is applied closely to the leaf surface
