ChIP techniques usually start by the cross-link between DNA and proteins in a living cells using chemical cross-linkers or UV light. Subsquently, cross-linked chromatin is extracted and fragmented by digestion or sonication. The DNA–protein complexes under study are then immunoprecipitated using a specific antibody to the protein of interest and the obtained DNA is purified, released from the cross-link and analysed.
