After creating a library of oligonucleotides and incubating them with the target protein, the bound and free fractions are separated, usually through an EMSA, and the bound oligonucleotides are amplified by PCR, regarding DNA, or by RT-PCR succeeded by in vitro transcription, concerning RNA.
