Schematic representation of SpyTag and SpyCatcher fusion constructs for plant organelle targeting in Nicotiana benthamiana. All plant expression constructs are under control of the CaMV35S promoter and have a Kozak or Omega sequence in front of the start codon. For SpyTag fusion constructs, the organelle targeting sequence is at the N-terminus followed by a flexible glycine–serine linker (GSGSGSG), the SpyTag sequence and an HA-Tag at the C-terminus. Targeting sequences are OEP7 for chloroplastic (Plastid-SpyTag), and NtHxk1 for mitochondrial targeting (Mito-SpyTag). As reporter construct, eGFP was C-terminally fused to SpyCatcher, separated by glycine–serine linker and a C-terminal HA-Tag
