TALEN design for targeting MpNOP1. a Schematic depiction of left and right TALEN arms binding to a target site. Four basic 33–35 aa long variable repeats, depicted as blue (T), red (G), green (C), and orange (A) boxes, bind via their RVDs to the four distinct nucleotides. The color code corresponds to the binding sites of the TS1. N-termini of the left and right TALEN arms contain a NLS. For generation of the TALEN, the FokI endonuclease is fused to the C-termini of the TALE proteins.
