Isolated protoplasts were stained with 5-(and-6)-carboxy-2',7'-dichloro-fluorescein diacetate (CDCFDA) and lysed by osmotic swelling. The isolated protoplasts and induced vesicles were observed under light and epifluorescence microscopy.
