Miniprep DNA eluted from silica matrix at high concentration allows efficient transient gene expression assays. A, Cotyledon cells of Arabidopsis seedling expressing the WRKY29-GFP-MYC protein in the nucleus. Miniprep DNA (2 μg in 1.8 μl) was bombarded into 7-day-old Arabidopsis seedlings and the observation was made 12 h post biolistic bombardment. B, Arabidopsis mesophyll protoplasts expressing the WRKY29-GFP-MYC protein in the nucleus. Miniprep DNA (2.2 μg in 2 μl) was introduced into 4,000 Arabidopsis mesophyll protoplasts by PEG-mediated transfection and the observation was made 12 h after transfection. C, Maize mesophyll protoplasts expressing the WRKY29-GFP-MYC protein in the nucleus. Fifteen μg of miniprep DNA was used to transfect 106 maize protoplasts by electroporation and the observation was made 12 h after transfection. Scale bar = 20 μm.
