The tape sandwich method does not compromise expression patterns. A CFP-tagged ER marker (ABRC stock no.: CD3-953), the signal peptide ofAtWAK2 at the N-terminus of the FP, and the ER retention signal at C-terminus [8], was delivered into wild-type protoplasts by transient transfection (left). Protoplasts derived from stable transformation with the same plasmid (right, ABRC stock no.: CS16250) have the same expression pattern.
